Epigenetics defines heritable DNA modifications unrelated to DNA nucleotide sequence changes, affecting gene expression. These modifications may be heritable, but may also be caused by other factors such as age, the environment stimuli, lifestyle, and disease state. Epigenetic modifications include acetylation, methylation, phosphorylation, and ubiquitination and lead to changes in chromatin structure, which alter the accessibility of DNA to the transcription machinery and therefore influence gene expression. During tumor development, epigenetic changes including aberrant DNA methylation and histone modification in chromatin frequently occur. These epigenetic changes are potential targets for therapeutic intervention using inhibitors of DNA methylation and histone deacetylation.
